An efficient total synthesis of the potent V-ATPase inhibitor saliphenylhalamide (SaliPhe), a synthetic variant of the natural product salicylihalamide A (SaliA), has been accomplished aimed at facilitating the development of SaliPhe as an anticancer and antiviral agent. This new approach enabled facile access to derivatives for structure-activity relationship studies, leading to simplified analogs that maintain SaliPhe’s biological properties. These studies will provide a solid foundation for the continued evaluation of SaliPhe and analogs as potential anticancer and antiviral agents.

The complexation behavior, binding properties, and spectral parameters of supramolecular chirality induction in the achiral host molecule, syn (face-to-face conformation) ethane-bridged bis(zinc porphyrin), upon interaction with chiral bidentate guests (diamines and amino alcohols) have been studied by means of UV-vis, CD, fluorescence, 1H NMR, and ESI MS techniques. It was found that the guest structure plays a decisive role in the chirogenesis pathway. The majority of bidentate ligands (except those geometrically unsuitable) exhibit two major equilibria steps: the first guest ligation leading to formation of the 1:1 host-guest tweezer structure (K 1) and the second guest molecule ligation (K2) forming the anti bis-ligated species (1:2). The second ligation is much weaker (K 1 ? K2) due to the optimal geometry and stability of the 1:1 tweezer complex. The enhanced conformational stability of the tweezer complex ensures an efficient chirality transfer from the chiral guest to the achiral host, consequently inducing a remarkably high optical activity in the bis-porphyrin.

Enantioselectivity in the allylboration of W-silylimines with a variety of chirally modified allylboron reagents has been examined. Optically active N-sulfonylarhino alcohols (16,17 and 19) derived from D-camphor and norephedrine were found to be efficient chiral ligands for the allylboration reagent. These reagents smoothly reacted with Ar-silylimines to give the corresponding homoallylic amines in a high level of enantioselectivity up to 96% ee.

Synthetic Route of 23190-16-1, An article , which mentions 23190-16-1, molecular formula is C6H5CH(NH2)CH(C6H5)OH. The compound – (1R,2S)-(−)-2-Amino-1,2-diphenylethanol played an important role in people’s production and life.

(Chemical Equation Presented) A commercially available collection of beta-amino alcohols have been converted to their corresponding beta-hydroxy- and beta-(o-diphenylphosphino)benzoyloxy(o-diphenylphosphino)benzamides 11a-f and 12a-f and have been employed in the Tsuji-Trost asymmetric alkylation reaction with 1,3-diphenylpropenyl acetate. With the exception of ligands 11b and 11f, the beta-hydroxybenzoyloxy (o-diphenylphosphino)benzamide ligands 11a-f primarily afforded the (R)-enantiomer of the product. In contrast, the bis(phosphine) ligands 12a-f consistently afforded the (S)-enantiomer. The best ligand (12c) was derived from cis-(1R,2S)-2-amino-1,2-diphenyl-1-ethanol, and when applied in the asymmetric allylic alkylation reaction, it yielded the product in an enantiomeric ratio of 97.8.22 favoring the (S)-enantiomer. A computational study was conducted on the conformation that this ligand might adopt in the palladium-catalyzed alkylation reaction as compared to that of the Trost ligand 1a. 2009 American Chemical Society.

(Chemical Equation Presented) The first catalytic asymmetric Friedel-Crafts reaction of 2-methoxyfuran with nitroalkenes was developed under the catalysis of diphenylaminetethered bis(oxazoline)-Zn(OTf)2 complexes. The reaction conditions and ligands were optimized, and the scope of the reaction was tested by varying the nitroalkenes. For most of aromatic and heteroaromatic nitroalkenes, good yields and high enantioselectivities (86-96% ee) were obtained. The methoxyfuran group in the product can be transformed to carboxylic acid via oxidative fragmentation with full retention of the configuration.

Solvent-induced chirality switching in the optical resolution of racemic tropic acid (TA) with (1R,2S)-2-amino-1,2-diphenylethanol has been demonstrated. Recrystallization of the diastereomeric salt mixture from i-PrOH or EtOH afforded the (S)-TA salt, while the (R)-TA salt was deposited from 1,4-dioxane and water-enriched alcohol solutions. Dual chirality switching was achieved by using two different types of solvents. The X-ray crystal structures of both diastereomeric salts showed that incorporation of the crystallization solvent played a crucial role in stabilizing each diastereomeric salt crystal. The mechanism of chirality switching has been discussed on the basis of the relative stability of the salt, as deduced from their structures.
